Doxorubicin-Induced Cardiotoxicity: Mechanisms and Management

نویسندگان

چکیده

Unfortunately, anticancer medications are extremely harmful to normal cells. Doxorubicin (DOX) is a highly cardiotoxic medication that can result in cardiomyopathy. The most significant mechanism for DOX-induced cardiotoxicity oxidative stress, while other pathways have also been put forth. This review aims highlight the mechanisms of and updated managements. Trustworthy sites such as Google Scholar, PubMed, Research Gate were used find articles. Many titles searching, "doxorubicin cardiotoxicity," "cardioprotection," "cardio-protective effects phytochemicals." Preprints, articles, research with meta-analyses disregarded. Three pathways, including mitochondrial damage, calcium excess, responsible cardiotoxicity. Cardiotoxicity may be partially caused by cell death, activation ubiquitin-ligase-proteasome system, changes its gene expression brought on DOX. In instance DOX cardiotoxicity, nutraceuticals antioxidants iron chelating properties found cardio-protective benefits. conclusion, doxorubicin-treated cancer patients linked making cardioprotection important these patients. All included this review's discussion might provide light fresh approaches treatment and/or prevention

Doxorubicin Induced Cardiotoxicity: Possible Cause and Preventions

Doxorubicin/Adriamycin (Dox) is a powerful, well established and highly efficacious drug in the fight against many kinds of cancers like solid tumors, leukemia’s, soft tissue sarcoma, breast cancer, small cell carcinoma of the lung and esophageal carcinomas. But its clinical usefulness is still restricted due to its specific toxicities to cardiac tissues. Congestive heart failure, cardiomyopath...

Low dose radiation prevents doxorubicin-induced cardiotoxicity

This study aimed to develop a novel and non-invasive approach, low-dose radiation (LDR, 75 mGy X-rays), to prevent doxorubicin (DOX)-induced cardiotoxicity. BALB/c mice were randomly divided into five groups, Control, LDR (a single exposure), Sham (treated same as LDR group except for irradiation), DOX (a single intraperitoneal injection of DOX at 7.5 mg/kg), and LDR/DOX (received LDR and 72 h ...

Transcriptional analysis of doxorubicin-induced cardiotoxicity.

Doxorubicin is an effective chemotherapeutic agent against a broad range of tumors. However, a threshold dose of doxorubicin causes an unacceptably high incidence of heart failure and limits its clinical utility. We have established two models of doxorubicin cardiotoxicity in mice: 1) in an acute model, mice are treated with 15 mg/kg of doxorubicin once; and 2) in a chronic model, they receive ...

Neuregulin1-ErbB Signaling in Doxorubicin-Induced Cardiotoxicity

This chapter will review basic and clinical findings regarding the cardioprotective role of Neuregulin1-ErbB signaling against the cardiotoxicity of doxorubicin, a widely used chemotherapeutic agent. In 2001, The New England Journal of Medicine published the results from clinical trials in breast cancer patients using Trastuzumab, a monoclonal antibody that blocks the ErbB2 receptor. These stud...

UVRAG Deficiency Exacerbates Doxorubicin-Induced Cardiotoxicity

Doxorubicin (DOX) is an effective chemotherapeutic drug in the treatment of various types of cancers. However, its clinical application has been largely limited by potential development of cardiotoxicity. Previously we have shown that ultra-violet radiation resistance-associated gene (UVRAG), an autophagy-related protein, is essential for the maintenance of autophagic flux in the heart under ph...
